Here are the discerning features of the two:

Both modify the machine tool kinematics model.

Both share the same user interface, called the Machine Tool Navigator.

You can activate the Machine Tool Builder from the NX application menu, and it is independent of the Manufacturing application. Use it to edit the kinematics in the machine model part file which resides in the file management of the machine library. The system copies the kinematics model into the manufacturing part when it retrieves the machine from the library. Thus, modifications of the kinematics in the machine part file are not automatically reflected in all the manufacturing parts that make use of the machine. You have to edit the manufacturing parts and retrieve the machine again.

The Setup Configurator is part of the Manufacturing application and modifies the copy of the machine kinematics model in the manufacturing part. Use it to add parts and fixtures to a machine setup.

Although it is possible to modify the kinematic structure of the machine using the Setup Configurator, it is important to keep in mind that any changes made here will only affect the CAM part. And all these changes will be lost if you should do a replace machine.

Once you have initialized the manufacturing environment in a part (entered the Manufacturing application), you can no longer modify the part from the MTB, and it can no longer become a library machine.
